阿利新蓝(Alcian)又称爱先蓝或阿尔辛蓝等,是一种类铜钛花青共轭染料,最初用于纺织纤维染色。这种阳离子染料与酸性基团结合,也即阿尔辛蓝与组织内含有的阴离子基团如羧基和硫酸根形成不溶性复合物。阿利新蓝由中央含铜的酞菁环与四个异硫脲基通过硫醚键相连而成。该异硫脲基呈中度碱性,使阿利新蓝带阳离子。阿利新蓝使碳水化合物着色的确切机制不明, 普遍认为是阳离子的异硫脲基通过静电与组织内的多聚阴离子相连。如含羧基和硫酸根的酸性黏液物质的羧基和硫酸根形成不溶性复合物,即染料分子中带正电荷的盐键和酸性黏液物质中带负电荷的酸性基团结合呈蓝色。
该试剂盒主要由Alcian 染色液和复染液组成,适用于细胞样本染色。染色结果为:酸性黏蛋白呈蓝色,细胞核呈红色。

您好,该染色液可以同时对糖胺聚糖和糖蛋白进行染色,但是糖胺聚糖易溶于水需要使用甲醇固定,糖蛋白可以使用多聚甲醛室温固定30min替代甲醇固定。对于六孔板建议每孔添加1ml。
