Lymphocyte（淋巴细胞，2.5 μg/ml ConA ）

Purified lymphocytes were collected by centrifugation (1800 r,10 min). Lymphocytes were suspended with RPMI 1640 medium containing 4% fetal bovine serum, 2.5 μg/ml ConA (Beijing Solarbio Science Technology Co., Ltd., Beijing, China) and seeded in 6-well plates at a density of 2 × 106 cells/mL

T/B lymphocyte（T淋巴细胞，4µg/ml Con A）

The proliferation of spleen T/B lymphocyte was also determined using an MTT assay as above. Briefly, 100 µl of cell suspension was seeded in a well of 96-well culture plates. After the addition of 50 µl Con A (16 µg/mL) or LPS (4 µg/mL) and 50µL COS-Se (80, 200, 400, 800 and 2000 µg/mL), the plates were incubated for 48 h at 37°C in a 5% humidified CO2. The stimulation index (SI/%) was calculated using the following equation: 

SI (%) = Absorbance of COS-Se groups/Absorbance of Con A or LPS groups × 100%.

Lymphocyte（淋巴细胞，10 µg/mL ConA）

Within 24 h of the last administration, the mice were sacrificed in a sterile environment. Spleen tissue was disrupted, and spleen cell suspensions were passed through sterile nylon mesh. Red blood cells were lysed by Erythrocyte Lysate (Solarbio, China). Cells were resuspended in RPMI 1640 medium (Solarbio, China) at the concentration of 5 x 106 cells/L. Blastogenic response of splenocytes to the mitogens, concanavalin A (ConA, Solarbio, China), and lipopolysaccharide (LPS, Solarbio, China), were assessed by CCK-8. Splenocytes suspension was incubated with ConA (10 µg/mL) or LPS (15 µg/mL) in 150µL RPMI 1640 medium containing 10% foetal bovine serum at 37 °C with 5% CO2. After incubation for 44 h, 10 lL CCK-8 was added to each well. After incubation for 2 h, the absorbance at 450 nm was measured by a microplate reader.

In Vivo

Mice（Female C57BL/6 mice (6-week-old)，15 mg/kg ConA ，iv. ）

All the mice were acclimatized for 1 week and then randomized into four groups (n=6-7 per group). According to the previous studies mice were intravenously (i.v.) injected with ConA (15 mg/kg; Solarbio, Beijing, China) for 12 h to establish a mouse model of IMH.

In Vitro( mouse lymphocyte )

The separated lymphocytes were cultured with a final concentration of 0, 24.57 ± 0.55, and 100.56 ± 1.48 μg/mL LCNP or 1 μg/mL ConA for 48 h Subsequently, MTT was added, and the absorbance at 570 nm in each well was determined using a microplate reader.

Lymphocytes were seeded into 96-well plates at 5.23 ± 0.22 × 105 cells/well containing complete RPMI 1640 medium and treated with a concentration of 0, 24.57 ± 0.55, or 100.56 ± 1.48 μg/mL LCNP and 1.05 ± 0.13 μg/mL ConA.

Lymphocytes were cultured in a 24-well plate at 2.23 ± 0.28 × 106 cells/well with final concentrations of 0, 24.57 ± 0.55, or 100.56 ± 1.48 μg/mL LCNP and 1 μg/mL ConA.
