测定意义：乙酰辅酶A 广泛存在于动物、植物、微生物和培养细胞中。是生物体能源物质代谢过程中产生的一种重要的中间代谢产物。在体内能源物质代谢中是一个枢纽性的物质。糖、脂肪、蛋白质三大营养物质通过乙酰辅酶A 汇聚成一条共同的代谢通路-三羧酸循环和氧化磷酸化，经过这条通路彻底氧化生成二氧化碳和水，释放能量用于ATP 合成。此外，乙酰辅酶A 是合成脂肪酸，酮体，胆固醇及其衍生物等生理活性物质的前体物质。
测定原理：苹果酸脱氢酶可催化苹果酸和NAD 生成草酰乙酸和NADH。柠檬酸合酶可催化乙酰辅酶A和草酰乙酸生成柠檬酸和辅酶A。利用苹果酸脱氢酶和柠檬酸合酶的偶联反应，乙酰辅酶A含量和NADH 的生成速率成正比，340nm 下吸光值的上升速率反应了乙酰辅酶A 含量的高低。
需自备的仪器和用品：紫外分光光度计、水浴锅、台式离心机、可调式移液器、1 mL 石英比色皿、研钵、冰和蒸馏水。
