In Vitro

Western blotting. （1 µM Leupeptin）

MCF-7 cells were treated with the aforementioned CoCl2 concentrations for 24 h and cells were lysed with ice-cold radioimmunoprecipitation assay buffer (150 mM NaCl, 1.0% NP-40, 0.1% Triton X-100, 0.5% sodium deoxycholate, 0.1% SDS and 50 mM Tris-HCl, pH 8.0) and protease inhibitors (AEBSF at 2 mM, Aprotinin at 0.3 µM, Bestatin at 116 µM, E-64 at 14 µM, Leupeptin at 1 µM and EDTA at 1 mM; Beijing Solarbio Science & Technology Co., Ltd., Beijing, China).

Western Blotting（0.5 ug/mL Leupeptin）

The mice heart (n=3) was dissected out and homogenized with a homogenizer at 4 °C in ice-cold lysis buffer containing 150 mM NaCl, 50mM Tris (pH 7.4), 1% sodium deoxycholate, 1%TritonX-100, 0.1% SDS, 25 mM β-glycerophosphate, 2m M sodium pyrophosphate, 1mM EDTA, 1mM Na3VO4 and 0.5 ug/ml leupeptin (Solarbio, Beijing, China).  The homogenate was centrifuged at 12,000×g for ten minutes at 4 ℃ to obtain the supernatant which was used to analyze the protein expression.

Cell（HEK293T cells和 hESCs，1 μg/mL Leupeptin，30 min at 4◦C on a rotation wheel）

For TFAP2C-OTX2 Co-immunoprecipitation, HEK293T cells expressing either HA-tagged OTX2 and MYC-tagged TFAP2C protein were collected after being scraped, pelleted, and washed three times using 1 × PBS. The cells were lysed with RIPA buffer supplemented with a cocktail of protease inhibitors (1 μg/mL Aprotinin (Solarbio, IA0430), 1 μg/mL Leupeptin (Solarbio, IL0070), 1 μg/mL Pepstatin (Solarbio, IP0170) and 1 mM PMSF) for 30 min at 4◦C on a rotation wheel. The soluble cell lysates were collected after maximum speed centrifugation at 4◦C for 30 min. Subsequently, 1 μg/mL of HA beads was incubated with cell lysates for 2 hrs at 4◦C on a rotation wheel. Beads were then washed for three times with PBS. Proteins were eluted in 1 × SDS loading buffer for subsequent Western Blot analysis. For MacroH2A.1-OTX2, MacroH2A.1-TFAP2C Co-immunoprecipitation, control hESCs expressing either 3 × FLAG-tagged OTX2 and MacroH2A.1 or control and OTX2-KO hESCs expressing 3 × FLAG-tagged OTX2 and MacroH2A.1 protein were collected after being scraped, pelleted, and washed three times using 1 × PBS. The cells were lysed with RIPA buffer supplemented with a cocktail of protease inhibitors (1 μg/mL Aprotinin (Solarbio, IA0430), 1 μg/mL Leupeptin (Solarbio, IL0070), 1 μg/mL Pepstatin (Solarbio, IP0170) and 1 mM PMSF) for 30 min at 4◦C on a rotation wheel. The soluble cell lysates were collected after maximum speed centrifugation at 4◦C for 30 min. Subsequently, 1 μg/mL of FLAG antibody with beads and 1 μg/mL of MacroH2A.1 antibody with beads were incubated with cell lysates for 2 hrs at 4◦C on a rotation wheel. Beads were then washed for three times with PBS. Proteins were eluted in 1 × SDS loading buffer for subsequent Western Blot analysis.
