UPLC-MS

Ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) was performed with liquid mass spectrometer for evaluation. The determination was performed on the SB-C18 column at the flow rate of 0.3 mL/min and the column temperature was 40 ◦C. The flavonoid and anthocyanin components were detected at 210 nm.

HPLC

HPLC was performed using an equipped with a DAD detector and a C18 (250 mm × 4.6 mm; 5 µm). The mobile phase included 0.2% phosphoric acid plus 0.4% sodium 1-heptanesulfonate (A) and methanol (B) at a flow speed of 1.0 ml/min in the condition of a column temperature of 30℃. The detection wavelength was set at 214 nm. The reference standard sample injection volume was 10 µl. The gradient elution was as follows: 0–5 min (100–100% A, 0%–0% B), 5–60 min (100–60% A, 0%–40% B), 60–120 min (60–30% A, 40–70% B), 120–125 min (30–30% A, 70–70% B), 125–125.1 min (30–100% A, 70–0% B), 125.1–132 min (100–100% A, 0–0% B). The chromatographic data and peak area scores were collected and analyzed using ChemStation software.

UPLC-QTOT-MS

The C18 (2.1 × 100 mm, 1.7 µm) was kept at 30 ℃. The injection volume was 1 µL, and the elution was completed in 18 min with a flow rate of 0.3 mL/min. Solvents A (water + 0.1% formic acid) and B (acetonitrile) were used in the following gradients: 0–2 min (5–10% B), 2–10 min (10–20% B), 10–15 min (20–40% B), 15–17 min (40–70% B), and 17–18 min (70–100% B). The PDA spectra for phenolic compounds were measured at 320 nm and 350 nm. For MS analysis, anthocyanins were analyzed in the positive ion (PI) mode; other phenolic compounds were analyzed in the negative ion (NI) modes. The MS parameters were as follows: source temperature of 120 ℃, desolvation temperature of 250 ℃(400℃ in the positive ion), cone gas flow 50 L/h, desolvation gas flow 600 L/h (800 L/h in the positive ion), source capillary of 3.0 kV. The MS analysis was performed using mass scanning from m/z 50 to 1,500.

LC–MS/MS

Chromatographic separation was achieved on a C18 Column (150 mm × 4.6 mm, 5 m). The mobile phase contained 0.1% formic acid aqueous solution (solvent C) and acetonitrile (solvent D) with a flow rate of 0.8 mL/min. The column temperature was maintained at 25 ◦C. The injection volume was 10 L. The chromatographic separation was conducted by the following gradient program: 0-0.5 min (30-30% B), 0.5-6 min (30–85% B), 6-8 min (85-92% B),8-10 min (92-95% B), 10-10.1 min (95-30% B), 10.1-12 min (30%B)

All analytes were detected utilizing turbo ionspray ionization with the following mass spectrometer source settings: ion spray voltage, -4.5 kV; the turbo spray temperature, 650℃; nebulizer gas (gas1), 60 L/min; heater gas (gas2), 65 L/min; curtain gas, 35 L/min. The selected precursor/production pairs were 343.0→328.2 for nevadensin, 359.2→161.0 for rosemary acid, 521.0→323.2 for salviaflaside, 178.9→134.9 for caffeic acid, 301.0→150.9 for quercetin, 285.0→132.9 for luteolin and 252.2→155.9 for sulfamethoxazole.
