铁是人体必需的微量元素之一，对于维持机体正常生理功能具有重要作用。亚铁离子是血红蛋白、肌红蛋白、细胞色素及其他酶系统的重要组成成分，帮助氧的运输，促进脂肪氧化。缺乏铁元素容易造成贫血、代谢紊乱，并影响机体的免疫功能。铁过量是引发或加剧多种慢性病（如糖尿病、心脑血管疾病、神经退化性疾病等）的危险因素。
样本中的Fe2+在酸性条件下与三吡啶基三嗪形成蓝色配合物，在593nm处有吸收峰，通过测定该波长吸光度即可计算Fe2+的含量。
