琼脂粉是由琼脂糖（agarose）和琼脂果胶（agaropectin）组成的，溶解性是琼脂粉不溶于冷水，易溶于沸水，缓溶于热水。检测凝胶强度为1.5%琼脂粉溶液在20摄氏度凝固后，能够表面不破承受的最大压强是800-1200g/cm2 在生物学实验中，微生物培养基、植物组培培养基常规添加琼脂粉的用量1.5-2% （工作浓度6-25g/L）。当实验中遇到琼脂粉不凝固时，常规的检测方法时pH值，浓度，温度，充分混匀等情况。
说明：培养基的配制，工作浓度6-25g/L
用途：微生物培养基、植物培养基原材料，一般添加量1.5-2%
琼脂粉默认使用PH为中性， PH小于4则无法凝结，
脂粉质检凝胶强度为1.5%浓度，800-1200g/cm2
常见问题解答：
一，生物实验中遇到培养基加入琼脂粉后，室温不凝固的原因分析：
1、培养基PH值过低 ，偏酸，水解，琼脂粉在PH值小于4则无法凝结。
2、所加琼脂粉质量不够 ，增加用量。搅拌不均匀，倒板前没有充分摇匀，凝固不均匀。
3、灭菌温度过高，加热时间过长。PH改变或破坏了琼脂的凝固度
4、培养基没有加热溶解充分，就没有凝固。
二，琼脂粉、琼脂糖、低熔点琼脂糖在生物实验中的区别分类：
琼脂粉是由琼脂糖（agarose）和琼脂果胶（agaropectin）组成的。琼脂糖是线性的多聚物，基本结构是1,3连结的β-D-半乳呋喃糖和1,4连结的3,6-脱水α-L-半乳呋喃糖。琼脂果胶是由许多更小的分子组成的异质混合物。它们的结构相似，但带硫酸根和羧基组分，凝胶能力差。简单地说，把琼脂中的琼脂果胶去掉，剩下的部分，就是琼脂糖。
在工业上的琼脂色泽由白到微黄，具有胶质感，无气味或有轻微的特征性气味，琼脂不溶于冷水，易溶于沸水，缓溶于热水。
琼脂糖在水中一般加热到90℃以上溶解，温度下降到35-40℃时形成良好的半固体状的凝胶，这是它具有多种用途的主要特征和基础。琼脂糖凝胶性能通常用凝胶强度表示。强度越高，凝胶性能越好。质量较好的琼脂糖强度通常在1200克/cm2以上（1%胶浓度）。琼脂糖的凝胶性是由存在的氢键所致，凡是能破坏氢键的因素都能导致凝胶性的破坏。琼脂糖具有亲水性，并几乎完全不存在带电基团，对敏感的生物大分子极少引起变性和吸附，是理想的惰性载体。在琼脂糖制备过程中需要把琼脂果胶尽量去除，否则琼脂糖有可能存在极微量硫酸根和丙酮酸取代电离基团，就会造成电内渗（EEO），电内渗对质点的移动产生影响。质量较好的琼脂糖硫酸根含量比较低，通常在0.2%以下，电内渗比较小，通常在0.13以下。这也就是琼脂糖比琼脂贵那么多的原因了。
问题1：使用琼脂粉配置的MS培养基颜色明显变黄？影响实验？
产品批次不一样，有些批次的琼脂粉就是显黄色，不是质量问题，不影响使用。
进口产品也有直接指出琼脂粉是黄色为正常现象。
问题2：凝胶强度？
答：凝胶强度（1.5%）≥800g/cm2 800-1200g/cm2，根据不同批次会有不同数值。
1.5g琼脂粉溶于100ml水，溶解凝固后要放置20℃恒温箱中静置10小时以上，再测定凝胶强度。与温度，水质等有关。
一般琼脂粉和卡拉胶，夏季使用浓度要比冬季偏高。夏季温度高，保存时间久，可能会有部分降解，所以要达到同样凝胶强度的用量就偏高。
问题3：琼脂粉不凝固
1、培养基PH值过低 ，偏酸，水解
2、所加琼脂量不够 ，增加用量。搅拌不均匀，倒板前没有充分摇匀，凝固不均匀。
3、琼脂质量不过关，琼脂条在使用时添加量要稍微高一点，琼脂粉是不是新买的
4、灭菌温度过高，加热时间过长。PH改变或破坏了琼脂的凝固度
5、培养基没有加热溶解够不够，就没有凝固。是不是琼脂加错了，营养琼脂不会凝固。
到平板时可以推荐在超净台内吹干晾干，蒸发过多水分。
问题4：PH与琼脂粉？
琼脂粉默认使用PH为中性，一般为7左右，PH小于4则无法凝结，具体的PH与稳定性的检测没有做过，根据经验来说 PH偏碱，凝胶后颜色稍白，PH偏酸，凝胶颜色稍暗。
问题5：琼脂粉做碳源测试，不加碳源细菌仍然能生长，上一批次的就不能生长？
答：理论上应该不长，一般微生物都不能利用琼脂粉，但某些杂菌仍可以利用，所以要检测到底是琼脂粉纯度问题还是染了杂菌。
