S-UE能够水解尿素，产生氨和碳酸。土壤脲酶活性与土壤的微生物数量、有机物质含量、全氮和速效氮含量呈正相关。土壤脲酶活性反应了土壤的氮素状况。
本法以尿素为基质，利用靛酚蓝比色法测定脲酶水解尿素产生的NH3-N，生成的蓝色靛酚和氨的浓度成正比。
