UHPLC-Q-TOF

The analysis of metabolites was carried out using an UHPLC LC 30 for the chromatographic determination. The extract was injected into a C18 analytical column (2.0 mm I.D × 75 mm, 1.6 μm particle size). The mobile phase was 0.1% formic acid solution (solvent A) and acetonitrile (solvent B). The gradient elution programmer was as follows: 0~2 min, 2% B;2~13 min, 2–90% B;13~16 min, 100% B;16~17 min, 100-2% B;17–20min, 2% B. The flow rate was of 0.3 mL/min, and the column temperature was set at 40 ℃. The injection volume is 3 μL. The peak detection of the extract was used a X500R Q-TOF mass detector. Both primary mass and secondary fragmentation pattern (MS/ MS) spectra were recorded between m/z 50 and m/z 1000 Da.

HILIC-UHPLC–QTRAP®/MS2

All NTs were separated on an Acquity UPLC BEH amide column (2.1 mm × 100 mm, 1.7 µm) equipped with an Acquity UPLC BEH Amide VanGuard Pre-column. The mobile phase was A (0.2% formic acid-5 mmol⋅L-1 ammonium formate and ammonium acetate water solution) and B (0.2% formic acid-1 mmol⋅L-1 ammonium formate and ammonium acetate water solution). The gradient elution was 0–3.0 min, 85–80% B;3.0–4.0 min, 80–54% B;4.0–4.5 min, 54–54% B;4.5–5.0 min, 54–85% B;and 5.0–6.0 min, 85–85% B. The equilibration time was 1 min, and the flow rate of mobile phase was set at 0.4 mL⋅min−1 with a sample injection volume of 2 µL. The column temperature was set at 30 ℃.

LC-MS/MS

LC-MS/MS conditions Analysis was performed using a liquid chromatographytandem mass spectrometer LC–MS/MS equipped with an ESI source. Separation was performed using a C18 column (250 mm × 4.6 mm × 5 μm). The flow rate was 0.4 ml/min, and the column temperature was maintained at 25℃. The mobile phases were formic acid: water (0.1:100, v/v) as mobile phase A, and methanol:acetonitrile (1:1) and 0.1% formic acid as mobile phase B. The binary gradient elution conditions were: (A:B): 0.01 min–5 min, 70:30→5:95;5–8 min, 5:95;8.01 min–16 min, 70:30. Detection was performed using multiple reaction monitoring (MRM) in positive mode, and the optimized MRM parameters for each compound are shown in Table 1. The mass condition parameters were set as: nebulizer gas, 3 L/min;drying gas, 10.0 L/min;heating gas, 10.0 L/min;interface temperature, 30℃;collision-induced dissociation (CID) gas, 230 kPa;DL temperature, 250℃;thermal block temperature, 400℃;interface voltage, −4.5 kV. The autosampler was kept at 4℃.
