Caspase-3是凋亡过程中最主要的终末剪切酶，因而也是研究最多的caspase，它激活pro-caspase-2,6,7,9等，特异水解多种凋亡关键蛋白如PARP，介导细胞核染色质固缩、凋亡小体生成、核DNA断裂。试剂盒测定原理基于Caspase-3特异水解多肽底物acetyl-Asp-Glu-Val-Asp p-nitroanilide (Ac-DEVD-pNA)，释放硝基苯胺pNA。游离硝基苯胺pNA呈黄色在405 nm具有最大吸收峰，用普通可见光分光光度比色方法测定。根据pNA标准管吸光度OD值和标准曲线，计算来自底物肽释放的pNA浓度，得到Caspase-3水解活性。反应体系100微升，试剂盒提供的试剂，除标准曲线外可进行50或100次样品测定。
适用：测定哺乳动物组织、细胞caspase-3活性。
组成与储存：
1. 裂解缓冲液： 20 ml ，4 ºC
2. 反应缓冲液： 10 ml ，4 ºC
3. 2 mM Ac-DEVD-pNA底物： 0.5 ml，－20 ºC避光
4. 10 mM pNA标准品： 0.5 ml，－20 ºC避光
以上标出的量为100次检测，50次检测的试剂量减半。试剂常温运输。到达后按要求储存，1年内稳定。
所需仪器：
酶标仪与96孔酶标板；或可见光分光光度计配100μl容积的石英比色杯。
工作波长：最大吸收波长405 nm，如不具备也可在400~450 nm范围内测定。
