高密度脂蛋白(High-Density Lipoprotein Cholesterol,HDL-C)是血清脂蛋白中密度最大、颗粒最小的脂蛋白,主要作用为将周围组织的胆固醇运送到肝脏进行降解。众多流行病学研究表明,HDL-C水平与动脉粥样硬化(AS)、冠心病(GHD)呈负相关,对于临床诊断动脉粥硬化、冠心病、高血压等疾病有重要参考价值。
使用选择性沉淀剂将HDL-C特异性分离出来,利用酯酶催化胆固醇酯水解生成游离胆固醇(FC)和游离脂肪酸(FFA),从而把胆固醇酯转化为FC;进一步利用胆固醇氧化酶催化FC氧化,生成4-胆甾烯酮和H2O2;最后利用过氧化物酶催化H2O2氧化4-氨基安替比林和苯胺类似物,生成紫色化合物,其在546nm有特征吸收峰,其颜色深浅与HDL-C含量成正比。
