真菌是具有真核和细胞壁的异养生物，种属很多，已报道的属达1万以上，种超过10万个。真菌通常又分为三类，即酵母菌、霉菌和蕈菌（大型真菌）。对于酵母菌和霉菌，可以用玻璃珠处理，而对于大型真菌，可直接用液氮研磨。经过前期处理的菌液，用硅质膜吸附，即可得到高纯度的基因组。提取纯化后的DNA，可以直接用于 PCR/Real time-PCR，sequencing，Southern blot，mutant analysis，SNP 等下游应用实验。

您好，试剂盒过期后不建议使用，当然也可以用少量菌做个预实验看看DNA提取效率以及纯度能不能符合要求。
