低密度脂蛋白是血清脂蛋白中胆固醇含量最高的一种脂蛋白,其颗粒较小,主要作用是将肝脏组织的胆固醇经过血液转运至其他组织,促进组织细胞中胆固醇的积累。众多流行病学研究表明,血清低密度脂蛋白胆固醇(Low-Density Lipoprotein Cholesterol,LDL-C)水平与动脉粥样硬化(AS)、冠心病(GHD)呈正相关,对于临床诊断动脉粥硬化、冠心病、高血压等疾病有重要参考价值。
使用选择性表面活性剂,特异性解离出CM、VLDL、HDL中的胆固醇,而不作用于LDL,解离出的胆固醇与胆固醇酯酶(CHER)和胆固醇氧化酶(CHOD)反应产生H2O2,在缺乏显色剂的情况下被消耗掉而不显色;未被分解的LDL在另一特异性表面活性剂的作用下解离,利用酯酶催化胆固醇酯水解生成游离胆固醇(FC)和游离脂肪酸(FFA),从而把胆固醇酯转化为FC;进一步利用胆固醇氧化酶催化FC氧化,生成4-胆甾烯酮和H2O2;最后利用过氧化物酶催化H2O2氧化4-氨基安替比林和苯胺类似物,生成紫色化合物,其在546nm有特征吸收峰。
