产品简介：
NBT即氯化硝基四氮唑兰 (Nitroblue tetrazolium chloride)，为深蓝色无定形微溶物质。BCIP即 5-溴-4-氯-3- 吲哚基-磷酸盐(5-Bromo-4-Chloro-3-Indolyl Phosphate), 在碱性磷酸酶（Alkaline phosphatase,AP）的催化作用下，
BCIP会被水解产生强反应性的产物，该产物会和NBT发生反应，形成不溶性的深蓝色至蓝紫色的沉淀。
使用说明：
1. 取 40ul 25×NBT 加入到 1ml 1×AP 反应缓冲液中，混匀后，再加入 40ul 25×BCIP 混匀，即配成反应工 作液，此反应液最好现配现用。 注意：根据膜的大小，1×AP 反应缓冲液、25×BCIP、25×NBT 的量可以等比例扩大。
2. 把经洗涤的 PVDF/NC 膜浸入反应工作液中，于室温平缓摇动进行温育。
3. 当蛋白带的颜色深度达到要求（一般约 20-30 分钟）时，把膜浸入去离子水中停止反应。
注意事项：
BCIP、NBT 为可疑致癌物，请采取必要的防范措施。操作时应戴手套，尽量避免与皮肤接触，用后及时 彻底冲洗。

该试剂盒作用原理是：BCIP在碱性磷酸酶的催化作用下，BCIP 会被水解产生强反应性的产物，该产物会和NBT发生反应，形成不溶性的深蓝色至蓝紫色的沉淀。通常用于WB中作为AP标记抗体的显色底物，若直接染细胞中的碱性磷酸酶不确保显色底物有细胞膜通透性。需要染细胞中碱性磷酸酶试剂盒推荐使用我司货号为G1480的碱性磷酸酶染色试剂盒(偶氮偶联法)。
